Systems biology of signaling network

Cells respond to external stimuli and eventually make their decision for survival, growth and differentiation. In this process, the activities of signal transduction network and downstream transcription factors show dynamically rich, nonlinear behaviors. In cancer systems, this signal transduction network is often dysregulated by overexpression and mutation of genes in the network. To understand the relationship between the dynamics of signaling pathways and cell commitment, we analyze the network using mathematical modeling approaches. Our analysis indicated that there are several common rules to control cellular proliferation and differentiation. In particular, we identified threshold mechanisms for transcription factor activation which can possibly alter the cellular states digitally. In this seminar, I will introduce our modeling approaches for NF-κB signaling and ErbB receptor signaling network in immune and cancer cells, respectively.
